QuEChERS 方法对茶叶中多种农药残留的 UPLC-MS/MS 测定

一、样品提取
准确称取 2.0 g 经粉碎的绿茶于 50 mL 离心管中,加 10 mL 水涡旋混匀 30s,静置 30 min,加入 15 mL 1% 乙酸乙腈,2500 rpm 涡旋 2 min,然后加入 6 g 无水硫酸镁,1.5 g 醋酸钠盐包(Cat.No.COQ050020H), 手动混匀后 2500 rpm 涡旋5 min,5000 r/min 离心5 min,上层乙腈层待净化。
二、样品净化
取 8 mL 待净化液加入至 15 mL QuEChERS 净化管(Cat. No.COQ015047H),涡旋 5 min,5000 r/min 离心 5 min, 准确吸取 2 mL 上清液于另一干净 15 mL 离心管中,40℃水浴氮气吹至近干,用1 mL 60% 乙腈水溶液重新溶解,过0.22 µm 尼龙滤膜供上机测试。
三、标曲配制
称取空白茶叶样品 2.0 g 按照上述一、二步骤进行至氮吹近干,作为空白基质提取液;
分别精密量取一定量的混合标准液加入到空白基质提取液 中,用 60% 乙腈水溶液定容至 1 mL,配制成适当浓度的基质混合标准工作溶液。

仪器:UPLC-MS/MS(Thermo Fisher TQS Endura) 色谱柱:CommaSil® ODS (2.1 mm×100 mm,3.0 µm) 流动相:A:水(0.1% 甲酸)
B:甲醇(0.1% 甲酸) 洗脱方式:梯度洗脱,见表 1

六、注意事项
(1)该实验采用 UPLC-MS/MS 进行茶叶中多种农药残留的测定, QuEChER 方法净化,外标法定量,建议配制基质标曲进行定量, 否则会存在基质效应,影响仪器准确定量;
(2)从表 3 绿茶中多农残添加回收结果可知:大部分农药回 收率都在 70%-110% 之间,RSD 小于 10%,可以满足绝大多数客 户测试需求。但该法不适合做丁硫克百威,回收率 20% 以下, 是因为丁硫克百威在酸性条件下会分解为克百威,因此在酸性 条件下检测出克百威,还需调整方法重新验证。
(3)茶叶样本需先加水静置 30 min,因为干燥的茶叶是蜷缩 的,如采用此法直接用乙腈提取一次会造成提取不充分,结果 假阴性。因此需先加水使茶叶舒展,提高乙腈的提取效率, EN15662-2018 中对此也有详细的描述。
